To further qualify Lester's answer, you also have to go after keywords you have a chance of winning on. If a PR6 site holds the top notch, good luck on dethroning them. Also, you need to be careful of *where* you get your backlinks from, as Google also considers the quality of the site (the PR) from where they originate.

Simple google loves backlinks, hubpages is love by yahoo more than google, so you need to turn into social bookmarking sites like Digg.com the more backlinks and traffic you have the bigger your chance to be number 1 in google for any keyword.

In my experience there are three main factors to beconsidered:
1. Keyword competition. The lower the competition, the easier you get #1.
2. Site structure. I've build a mini site with just contains 5 pages (actually just 3 of them have content, since the other two is juts contact page and sitemap), and just by submitting about 5-6 link to social bookmarking and 1 hubs, I can get the First Page, outrank the other bigger site, But nit yet in #1.
3. Backlink. Quality backlink, this is what you need to push it to #1.
If you want my advice, go get the lower competition, and you'll see result soon. There's no use to go after the large competition where the all supersite with lots of staff sitting in first page of google.

Use Googel adword tools. Be sure who your competators are.. and your niche.

This is a big nut to crack but this technique has worked for me in the past, dethroning many top spots in less than a day. Not however, for a single word phrase, but give it a shot. Create a wordpress blog, on your own site, not the free version. Obviously it would be best if the phrase was in your domain name . Create a post whose title is SEO, place in a category called SEO and lastly make sure your permanlink is sitename.com/SEO/seo
Then go for your backlinks.
I have gotten number one this way without backlinks but I doubt it's likely to work without them for a single word phrase.
