Here's where you will find more movies like 500 Days of Summer. Its definitely difficult to find a movie that's similar to this one that doesn't fit easily any convenient genres.
I've included movies that I think captures some unique aspects of it while retaining their own individual flavor.
The movie 500 Days of Summer is not your typical romance movie. It looks at a whirlwind romance and breakup from the sometimes faulty and often selective memory of the nerdy guy.
The scene where they meet is one of the best boy meets girl of all time. It also sets the quirky tone for the rest of the movie. Amazingly this independently produced movies gets even better after the strong opening scene. It had me hooked waiting for the inevitable train wreck of the doomed relationship which we know in retrospect of course.

The Blu Ray version does wonders for the already fantastic soundtracks.
Boy Meets Girl
The boy in this case is Joseph Gordon-Levitt who falls head over heels in love with the girl, the talented singer/actress Zooey Deschanel.
So if you love movies by Joseph Gordon-Levitt then you might want to watch the one of the most mind bending movie by director Christopher Nolan in "Inception". For Miss Zooey Deschanel you can catch her in a smaller but interesting role as the damsel in distress in the spookfy fantasy adventure movie "Your Highness".

Dorky Guy + Killer Soundtracks = Garden State
If you haven't watched Garden State before then be prepared for one of the most neurotic & nerdy male protagonist ever.
This off beat sleep hit is also an independent production. Having thought about it, I don't think any sensible studio executive will have the fortitude to produced such a quirky gem.

Guy Who is a Humungous Failure + Meets Perky Girl While on the Way Home to Cremate Father = Elizabethtown
Take a guy that has incurred billions and billions of losses by producing the world's most hype failure of a sports shoe. Then add to it his father just died and he needs decide whether to cremate or bury him.
Then add a chance meeting with one perky sunshine of an air stewardess. A marathon soul sharing phone call. You now have a great movie that deals about failure, about the kindness of strangers and family, about how love can conquer all if you just let it in.
Oh and did I mention that it stars the Orlando Bloom and Kirsten Dunst. Need I say more.
