Maybe only on readers who choose to read the other comments. I think it shows that you are willing to hear both sides if you leave and respond to negative comments. (about the hub of course, if it's negative toward you as a person-that's a little different)
It won't hurt the score at all. If the person who left the comment happened to down rate your hub as well, that may cause the HubScore to drop a couple points, though

I read somewhere that it can affect your relationship with Google if people's comments breach Adsense TOS.
Yes, it's a good idea to preserve your Hubs and AdSense status by removing comments that violate HubPages or AdSense ToS.
You mean like bad language? Or race hate comments? Links to bad sites, etc...
Your best bet is to moderate all of your comments.
I have seen one word in a comment trigger PSAs on Blogs and no ads on HubPages.
