The best thing to do in the case of an automatic transmission is to go directly to the source, which would be inspecting the Automatic Transmission Fluid or ATF. Your ATF level and condition should be checked routinely. Improper fluid level can affect the transmission's behavior and lead to failure. The fluid should be a rosy red and not have a burned smell. The fluid should be topped off, but not overfilled, with the manufacturer's recommended fluid. Different makes have different fluid specifications. Your owners manual should have these specifications or have your local mechanic check for you.
Symptoms of a failing automatic transmission include slipping (the engine revs but the car doesn't move respectively), harsh or delayed gear engagement, harsh or delayed shifting or absence of any or all of the reverse or forward gears. Keep in mind that other problems within the power train may emulate a transmission problem.

As BigSeanR said, checking the fluid is probably the best way to diagnose transmission problems yourself. This should be done on a regular basis to spot problems before they become serious. Dark, burnt-smelling fluid is a sign that it needs to be changed and possibly inspected by a mechanic while a low fluid level could mean there is a leak. In this case, you should top up the fluid ASAP and if the problem continues, get your car inspected by a mechanic.
That said, there are a number of other warning signs including: lack of response, whining, clunking, humming, grinding, shaking, burning smell, doesn't go into gear, check engine light, doesn't go into gear, slipping and a dragging clutch. So I have a '92 t-bird and a 2006 taurus, both fords. I actually like the t-bird better than the taurus, except for the fact that the t-bird is in need of another transmission. I lost the overdrive while merging onto the freeway during busy hour, mileage was just barely 138,000...
